The church was built between 1520 and 1524 by the architect Antonio Medaglia and by will of the prince bishop Bernardo Clesio, on the remains of the medieval Paleo-Christian Church of Santa Maria ad Nives, in an area which returned numerous archaeological finds from its Roman past.

Inside there are remarkable artworks such as the organ choir by the sculptors Vincenzo and Girolamo Grandi from Vicenza, the altarpieces by Giovanni Battista Moroni, Pietro Ricchi, Francesco Unterperger and Giambettino Cignaroli and the organ doors – originally in the Dome - painted by Giovanni Maria Falconetto in 1508.
Starting from the 13th April 1562 the congregations of the third and final phase of the Council took place in this church. For this reason, a semi-circular wooden tribune was set in the nave, as it is proved by some paintings. In 1902 - to commemorate the Council's general congregations held here – the painter Sigismondo Nardi decorated the vault with scenes and characters from the Tridentine Synod.
